Microtubules are constituent parts of the mitotic appa- ratus, cilia, flagella, and elements of the cytoskeleton. They consist principally of 2 soluble proteins, alpha- and beta-tubulin, each of about 55,000 Da.Antibodies against beta Tubulin are useful as loading controls for Western Blotting. However it should be noted that levels of beta Tubulin may not be stable in certain cells.

IF image of BS00242-T stained A375 cells.The cells were 4% para- formaldehyde fixed (20 min) and then incubated in 10% normal goat serum for 1h to permeabilise the cells and block non-specific pro- tein-protein interactions. The cells were then incubated with the anti- body Tubulin β mAb (1:500) at 10µg/ml overnight at +4°C. The secondary antibody (Green) was Goat Anti-Mouse IgG (H+L) FITC#BS50950 used at a 1/400 dilution for 1h. Hoechst33342 #BD5011 was used to stain the cell nuclei (blue). |
